Question about Beckman 4.0 data analysis software


Dear colleagues,

We recently upgraded our Optima XL-A/XL-I data analysis software from
Origin version 3.78 to Origin version 4.0 from Beckman. The updated
version of Origin 4.0 is a much improved and user-friendly software.
However, in performing equilibrium analysis, when I imported data that
has been previously calculated with the older version, I obtain
different results with the newer software.  This could affect the
interpretation of data and change final experimental conclusions.  I
feel uneasy about this situation. I understand that simply editing of
the sedimentation equilibrium curve from experiment will yield some
difference, depending on how one makes the cut.  However, I did not
expect that the newer upgrade version would also introduce another
level of uncertainty.  My curiosity then lead me to calculate the
sample data set provided by Beckman's Origin 4.0, under
"C:\origin41\assoc", and compare the results obtained with each
versions (see attachment). As you can see from the result summary, the
% difference in molecular weight calculation is very good.  However the
Ka values are unacceptable for all but lower concentration sample
(20Ka.ra2).  This value of Ka in various concentrations of denaturing
solutions is very critical in order to understand the behavior of
different mutants in solution using the analytical ultracentrifuge
technique.
I hope that every RASMBers who has upgraded their data analysis
software to 4.0 can go through this exercise.  You can also try to
recalculate your old data using the newer version and compare the
results.
>From what I can tell, the self-association mathematical model used in
both versions is the same. Can anyone out there who is using Beckman's
software comment on this discrepancy?  I can accept the variation in
molecular weight, as well as association value for sample file 20Ka.ra2
(see attachment). But the discrepancy in association values for the
other files, including global fit, disturb me.
Since one obtains different results using two different version of
software, my question is whether or not we can say that the data
computed from the newer version (Origin 4.0) is more accurate than the
older version.  Can anyone comment on this? 
In February, I contacted Beckman so that this discrepancy would be
brought to their attention. However, I have yet to receive a response
from them.  I hope all the users of this updated version Origin 4.0
software be aware of this problem and check it out for yourself.  Thank
