Whilst Henryk Eisenberg's proposal is (obviously) totally correct in principle, it is 
necessary to bear in mind that the dialysis to equilibrium of a solution in 20% 
glycerol may take quite some time - possibly even a day or more with stirring.

And the protein concentration will surely change as a result of osmotic effects, so 
the protein concentration would need to be measured AFTER dialysis (always good 
practice, of course).  I suppose that if the extinction coefficient is well defined, then 
it will be OK to use it to estimate the concentration in the 20% glycerol (cannot 
think of any good reason why not, but have never actually done it !)
