Protein peptide binding

Hello Everyone;

        We are currently trying to study the binding of a peptide(Molecular weight 4000g/mole)
to a protein of molecular weight of 54,000 D,   I would like to get the openion of all of you 
in how should we approach this.  The binding constant of the peptide to the protein is not 
known,and the peptide bind to the protein in 1:1 stoichemitry. The molecular weight difference
between complexed and free protein is small to cause large change in the S value. Is it 
possible to get an association constant fromm equilibrium runs. If you have any suggestions or 
know of the work of anyone who work with similar systems please advise me. Your cooperation 
will be highly appreciated.
