Equilibrium Experiment

Hello;
    I'm trying to study the association of a protein that exist in a monomer
dimer equilibrium. The protein is stable and provided in a buffer that is 40%
glycerol, attempt to do equilibrium at the appropriate speeds has failed, and
no concentration gradient is established. I would like to get your input and
suggestion on whether it will it is possible to run the experiment under
certain conditions in the presence of this content of glycerol. Any suggestion
will be highly appreciated.                       Thanks for taking the time to
